a V-6 powered 2000 Mustang? I would think they would but I wanted to ask before I run out and buy a set. Has anyone bought a set of take-off wheels and tires from Saleen?

Absolutely.

That's exactly what I did too...bought a set of the take-offs from Saleen, and put 'em on my '99 V6. Couldn't beat the deal at the time...tires mounted on the wheels for $1K plus shipping...not a bad deal.
